Epitalon Handling Sheet
Also known as Epithalon, Epithalone, AEDG peptide
A tetrapeptide, Ala-Glu-Asp-Gly, whose Asp-Gly pair is one of the fastest-isomerizing motifs in peptide chemistry. Dry storage is straightforward; solutions need care.

Little compound-specific stability data is published. Guidance follows from the sequence and general peptide chemistry.
Bench card
- The Asp-Gly pair isomerizes readily in solution, forming isoaspartate.
- Keep solutions cold, slightly acidic and short-lived; freeze aliquots for anything longer.
- Acidic (net charge about −2 at neutral pH); dissolves easily in water.
- No oxidation-prone residues; light risk is low.
- Isomers have the same mass, so mass spectrometry alone will not detect them.
Handling-relevant features
| Feature | Why it matters for handling |
|---|---|
| Asp3-Gly4 | Classic fast isomerization site through a succinimide intermediate; produces isoaspartate |
| Glu2, Asp3, C-terminus | Acidic overall; very water-soluble |
| No aromatic or sulfur residues | Low oxidation and light risk |
What it is
Epitalon is a synthetic tetrapeptide, Ala-Glu-Asp-Gly, studied mainly by Khavinson and colleagues. Its literature can be found through PubMed.
The Asp-Gly site
An aspartate followed by glycine is one of the most reactive sites in peptide chemistry. Geiger and Clarke showed that aspartyl residues form a succinimide intermediate that reopens to give a mixture of normal aspartate and isoaspartate, and that the small glycine side chain makes ring formation especially easy. In Epitalon this site is a quarter of the molecule.
Storage
| Form | Recommended | Notes |
|---|---|---|
| Sealed powder | −20 °C | Desiccant; the dry solid is relatively robust |
| Solution, days | 2–8 °C | Slightly acidic; use promptly |
| Solution, longer | −20 °C or colder | Single-use aliquots |
